TelN Protelomerase 中文名称为 TelN 原核端粒酶，克隆自噬菌体 N15，其在识别位点 TelRL (56bp) 切割 dsDNA，TelRL 位点由中间的回文序列 TelO 和两端 14bp 的回文序列 R3、L3 构成 (Fig.1)，并在酶切位点处留下共价封闭的末端，从而有效地将环状 DNA 转化为有发夹末端的线性 DNA (Fig.2)。

酶活定义
在 50 μl 体系中，30°C 条件下,30 min 完全切割 0.5 μg 线性化 pMiniT-TelN 对照质粒(313 fmol TelN 识别位点)所需要的酶量定义为 1 个活力单位（U）。
产品使用步骤
（1）按照下表所示配制混合溶液：

（2）上下轻摇混匀体系，并短暂离心。
（3）30℃ 孵育 30 min。

TelN Protelomerase, whose Chinese name is TelN Prokaryotic telomerase, is cloned from phage N15, which cuts dsDNA at the recognition site TelRL (56bp), The TelRL site consists of a palindromic sequence TelO in the middle and a palindromic sequence R3 and L3 at both ends of 14bp (Fig.1), and leaves a covalently closed end at the cleavage site, thus effectively converting circular DNA into linear DNA with hairpin ends (Fig.2).

Enzyme Activity Definition
The amount of enzyme required to completely cut 0.5 μg linearized pMiniT-TelN control plasmid (313 fmol TelN recognition site) for 30 min in a 50 μl system at 30°C was defined as 1 unit of activity (U).
Protocol
(1) Configure the mixture as shown in the following table:

(2) Shake the mixed system gently up and down, and centrifuge briefly.
(3) Incubate at 30℃ for 30 min.
