产品简介

该试剂盒提供了一种与CWE2100匹配，从300 μl血液中提取DNA的预装型产品。样品按要求加入96 DW深孔板后，将其放入CWE2100中后约50分钟即可得到高质量的 DNA。提取得到的DNA稳定、可靠，可用于PCR、芯片检测，以及高通量测序等下游实验。

2 ．异丙醇（北京化工厂）

实验前准备及重要注意事项

1 ．向25 mg ProteinaseK中加入1.25 ml ProteinaseK Storage Buffer使其溶解，之后-20℃保存。配置好的ProteinaseK溶液勿长时间室温放置，避免反复冻融，以免影响其活性。

2 ．向装有Magbeads PN的试剂瓶中加入33 ml异丙醇（建议使用北京化工厂的异丙醇），混匀后备用。

3．使用前检查预装板中试剂是否存在漏液的情况。

操作步骤

1 ．将预装板从试剂盒中取出放入水平离心机中300×g离心10秒钟。

2 ．将深孔板从离心机中取出，小心去除热封膜，期间防止深孔板震动。

3 ．向深孔板的1&7列加入300 μl血液样品，之后向1&7列中加入20 μl Proteinase K。

4 ．将加入样品的深孔板放入仪器中，之后磁棒套固定在磁棒套架上。

5 ．运行Cowin Blood 300程序，约23分钟后仪器暂停。将深孔板从仪器中取出，向1&7 列中加入310 μl充分混匀的异丙醇与磁珠混合物。

6 ．将深孔板放回仪器中，继续运行程序。约35分钟后程序运行结束，取出磁套和深孔板。

7 ．将深孔板6&12列中的洗脱产物转移至1.5 ml离心管中低温保存。

Product Introduction
This reagent kit provides a match with CWE2100, ranging from 300 μ Pre packaged products for extracting DNA from blood. After adding the sample to the 96 DW deep well plate as required, it can be placed in CWE2100 for about 50 minutes to obtain high-quality DNA. The extracted DNA is stable and reliable, and can be used for downstream experiments such as PCR, chip detection, and high-throughput sequencing.

Preparation and important precautions before the experiment
1. Add 1.25 ml of ProteinaseK Storage Buffer to 25 mg of ProteinaseK to dissolve it, then maintain at -20 ℃ Save. The prepared ProteinaseK solution should not be left at room temperature for a long time to avoid repeated freeze-thaw cycles, which may affect its activity.
2. Add 33 ml of isopropanol (recommended to use isopropanol from Beijing Chemical Factory) to the reagent bottle containing Magheads PN, mix well, and set aside.
3. Before use, check if there is any leakage of reagents in the pre installed board.
Operation steps
1. Take out the pre installed plate from the reagent kit and centrifuge it in a horizontal centrifuge for 300 × g for 10 seconds.
2. Take out the deep hole plate from the centrifuge and carefully remove the heat sealing film to prevent vibration of the deep hole plate during the process.
3. Add 300 to columns 1&7 of the deep hole plate μ Add 20 to columns 1&7 of the blood sample μ Protein K.

4. Place the deep well plate containing the sample into the instrument, and then fix the magnetic rod sleeve onto the magnetic rod sleeve holder.
5. Run the Cowin Blood 300 program and the instrument will pause after approximately 23 minutes. Remove the deep hole plate from the instrument and add 310 to columns 1&7 μ Mix isopropanol and magnetic beads thoroughly.
6. Put the deep hole plate back into the instrument and continue running the program. After about 35 minutes, the program ends and the magnetic sleeve and deep hole plate are removed.
7. Transfer the elution products from columns 6 and 12 of the deep well plate to a 1.5 ml centrifuge tube for low-temperature storage.
