IF analysis of ACTA2 (green) in HeLa cells. The cells were fixed and permeabilized with 100% methanol for 5 minutes, and blocked with 2% BSA for 1 hour at room temperature. Cells were stained Recombinant ACTA2 Antibody (Ab088446) at 1/100 dilution in blocking buffer overnight at 4°C, and then incubated with Goat Anti-Rabbit IgG H&L (AF488) at a dilution of 1/1000 for 1 hour at room temperature in the dark (green). Cells were counterstained with DAPI (blue). Images were taken on the confocal laser scanning microscope.

The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% TritonX-100 for 15 min. The cells were then incubated in Recombinant ACTA2 Antibody (Ab088446) (1/100 dilution) in 1x PBS/1% BSA for 30 min at 4°C. The secondary antibody used was a Goat Anti-Rabbit AF488 (IgG H+L) at 1/2000 dilution for 20 min at 4°C. Unlabeled sample (Green) was used as a control.
